β-amiloide – fattore scatenante di malattie neuronali

I peptidi β-amiloidi sono derivati ​​dalla proteina precursore β-amiloide (APP) e, come placche amiloidi aggregate, sono un componente caratteristico di molte malattie neurodegenerative. Anche in piccole quantità, i peptidi sono tossici per le cellule nervose e, ad esempio nella malattia di Alzheimer, innescano la morte dei neuroni nel cervello. Le placche β-amiloidi sono costituite principalmente da due isoforme C-terminali del peptide con lo stesso nome, ciascuna con una dimensione di 40 (Aβ 1-40) e 42 (Aβ 1-42) aminoacidi vengono prodotte la secretasi β e γ.

Formazione di placche amiloidi

La scissione patologica della proteina precursore dell’APP ha come risultato che i peptidi β-amiloidi risultanti si assemblano in oligomeri e li organizzano come fibrille. Ciò si traduce in placche più grandi che causano reazioni infiammatorie, ostacolano la trasmissione sinaptica e interrompono il metabolismo neuronale, che alla fine porta alla morte delle cellule nervose.

Rilevazione di β-amiloide

Per una diagnosi esatta di varie malattie neurodegenerative, ad es. La malattia di Alzheimer, è essenziale essere in grado di identificare con precisione i componenti che sono responsabili dell’insorgenza della malattia. Mentre da un lato la proteina precursore dell’APP è presente in grandi quantità nel cervello umano e nel liquido spinale, il β-amiloide elaborato può essere misurato solo in quantità picomolari. Metodi comuni per la rilevazione di proteine ​​includono immunoistochimica, PCR quantitativa ed ELISA. Come per molte malattie dose-dipendenti, la diagnosi precoce è anche vantaggiosa per le placche amiloidi e può aiutare a contrastare il decorso della malattia.

In caso di malattia, le cellule della microglia del corpo cercano di imballare le placche di amiloide più strettamente al fine di prevenire l’aggiunta di nuovo β-amiloide. Varie opzioni di trattamento si concentrano principalmente sulla riduzione delle placche amiloidi nel cervello. Ad esempio, l’immunoterapia anti-amiloide potrebbe essere utilizzata per rimuovere le placche in modo mirato dopo la loro formazione.
